A high-dose of nerve growth factor (NGF) mixed with ginsenoside Rb₁(GRb₁) was encapsulated by collagen and placed in silicone rubber chambers, which were used to repair dissected Sprague-Dawley rat sciatic nerves with 15 mm gaps. Six weeks after surgery, no axons or Schwann cells were seen in these chambers. By comparison, nerves treated with collagen-GRb₁ alone had regenerated axons and Schwann cells in their endoneurial areas. We suggest that excessive NGF may not promote but, rather, suppress developing nerves.

The present study investigated whether Shenqi-wan possesses a protective effect against hydrogen peroxide (H₂O₂)-induced apoptosis of the hippocampal cell line HiB5. Through morphological and biochemical analyses, it was demonstrated that HiB5 cells treated with H₂O₂ exhibited several apoptotic features, while cells pre-treated with Shenqi-wan prior to H₂O₂ exposure showed a decrease in the occurrence of apoptosis. In addition, a patch clamp study revealed that Shenqi-wan inhibited profoundly N-methyl-D-aspartic acid (NMDA) receptor-activated ion current in acutely dissociated hippocampal CA1 neurons. These results suggest that Shenqi-wan may exert its protective effect against H₂O₂-induced apoptosis via inhibition of NMDA receptors in hippocampal neuronal cells.

In this study, hydrogen peroxide (H₂O₂)-mediated Caco-2 cytotoxicity was employed to investigate the potential antioxidant activity of the methanol extract from the lotus leaf (Nelumbo nucifera Gertn.). A dose-dependent protective effect against reactive oxygen species (ROS)-induced cytotoxicity was observed when Caco-2 cells were treated with 10 mM H₂O₂ in combination with the methanol extract of the lotus leaf (0.1–0.3 mg/ml). However, no significant effect was found when co-treating Caco-2 cells with 10 mM H₂O₂ and α-tocopherol. In vitro assay revealed that the extract exhibited scavenging activities on free radicals and hydroxyl radicals, and metal binding ability as well as reducing power, which may explain in part the mechanism behind the extract's ability to protect cells from oxidative damage. In addition, the extract also exhibited concentration-dependent antioxidant activities against hemoglobin-induced linoleic acid peroxidation and Fenton reaction-mediated plasmid DNA oxidation.

An ethanolic extract of Parsley, Petroselinum crispum (Mill.) Nym.ex A.W. Hill (Umbelliferae), was tested for its ability to inhibit gastric secretion and to protect gastric mucosa against the injuries caused by pyloric ligation, hypothermic restraint stress, indomethacin and cytodestructive agents (80% ethanol, 0.2 M NaOH and 25% NaCl) in rats. The extract in doses of 1 and 2 g/kg body weight had a significant antiulcerogenic activity on the models used. Besides, ethanol-induced depleted gastric wall mucus and non-protein sulfhydryl contents were replenished by pretreatment with Parsley extract. Acute toxicity tests showed a large margin of safety for the extract. The phytochemical screening of Parsley leaves revealed the presence of tannins, flavonoids, sterols and/or triterpenes.

Qing-Luo-Yin (QLY), a traditional Chinese herbal medicine for the treatment of rheumatoid arthritis, is a combination of the extracts of Sophora flavescens Ait., Phellodendron amurense Rupr., Sinomenium acutum Rehd. et Wils. and Dioscorea hypoglauca Palib. The suppressive effect of QLY on the development of angiogenesis was investigated in a rat model of collagen-induced arthritis (CIA). QLY (0.3 g/kg) was orally administered daily for 27 days. Neo-angiogenesis, pannus and cartilage damage, the expression of metalloproteinases (MMP)-3 messenger RNA (mRNA) and the level of the tissue inhibitor of matrix metalloproteinase (TIMP)-1 in the synovium were examined by histology, in situ hybridization and immunohistochemiscal assays, respectively. It was observed that the articular morphological alterations, the over-expression of MMP-3 mRNA and the reduced production of TIMP-1 in CIA rats were significantly ameliorated by QLY. QLY performed about as effectively as tripterygium glycosidorum tablets (0.1 g/kg) extracted from Tripterygium wilfordii Hook. f.. These results indicate that QLY exerts a suppressive effect on the angiogenesis of CIA rats, and suggest that the therapeutic effect of QLY could be due to restoring the balance of MMP-3 and TIMP-1 in rheumatoid synovium.

It has been reported that Nigella sativa oil possesses hepatoprotective effects in some models of liver toxicity. However, it is N. sativa seeds that are used in the treatment of liver ailments in folk medicine rather than its oil. Therefore, the aim of this study was to investigate the effect of the aqueous suspension of N. sativa on carbon tetrachloride (CCL₄)-induced liver damage. Aqueous suspension of the seeds was given orally at two dose levels (250 mg/kg and 500 mg/kg) for five days. CCL₄ (250 μl/kg intraperitoneally/day in olive oil) was given to the experimental group on days 4 and 5, while the control group was only treated with the vehicles. Animals treated with CCL₄ showed remarkable centrilobular fatty changes and moderate inflammatory infiltrate in the form of neutrophil and mononuclear cells when compared to the controls. This effect was significantly decreased in animals pretreated with N. sativa. Histopathological or biochemical changes were not evident following administration of N. sativa alone. Serum levels of aspartic transaminase (AST), and L-alanine aminotransferase (ALT) were slightly decreased while lactate dehydrogenase (LDH) was significantly increased in animals treated with CCL₄ when compared to the control group. LDH was restored to normal but ALT and AST levels were increased in animals pretreated with N. sativa. In conclusion, N. sativa seeds appeared to be safe and possibly protective against CCL₄-induced hepatotoxicity. However, further studies may still be needed prior to supporting its use in folk medicine for hepatic diseases.

The purpose of this study was to evaluate the effects of extracts of Coptidis rhizoma, Phellodendri cortex and Gardeniae fructus, which are medicinal herbs in Orengedoku-to (Huanglin-Jie-Du-Tang in Chinese), and crocetin (a major component of Gardeniae fructus) on experimental elevation of aqueous flare in pigmented rabbits. To produce aqueous flare elevation, 0.5 μg/kg lipopolysaccharide (LPS) was injected into the ear vein, or prostaglandin E₂ (PGE₂) 25 μg/ml, was applied to the cornea by means of a glass cylinder. Animals were pretreated by oral administration of 150 g/day of food containing 0.15% (w/w) extract powder of Coptidis rhizoma, 0.10% (w/w) extract powder of Phellodendri cortex or 0.15% (w/w) extract powder of Gardeniae fructus for 4 days, or by intravenous injection of crocetin, 0.3, 3, 30 or 300 μg/kg, 30 minutes before aqueous flare elevation. Aqueous flare was measured with a laser flare-cell meter. Aqueous flare intensity was expressed as the area under the curve (AUC) in arbitrary units. The AUC of LPS- and PGE₂-induced aqueous flare elevation was 4685 and 1386 arbitrary units, respectively. Pretreatment by oral administration of 0.15% (w/w) extract of Coptidis rhizoma or 0.10% (w/w) extract of Phellodendri cortex did not inhibit LPS-induced aqueous flare elevation. Pretreatment by oral administration of 0.15% extract of Gardeniae fructus suppressed LPS-induced aqueous flare elevation (AUC: 1411 arbitrary units). Pretreatment by intravenous injection of 3, 30 or 300 μg/kg of crocetin-inhibited LPS-induced aqueous flare elevation in a dose-dependent manner. Pretreatment with 3 or 30 μg/kg of crocetin did not inhibit PGE₂-induced aqueous flare elevation, but 300 μg/kg of crocetin inhibited PGE₂-induced aqueous flare elevation (AUC: 918 arbitrary units).

We investigated the effects of three traditional Chinese medicine prescriptions on changes of bone metabolism in mice, using a gravity device to produce a microgravity environment. We found that Hochu-ekki-to (TJ-41) and Hachimi-jio-gan (TJ-7) suppress the increase in the ratio of serum Ca/P and the increase of calcium in urine. Moreover, TJ-41 and Shin-bu-to (TJ-30) reversed the increase of alkaline phosphatase activity (ALP), and TJ-41 also reversed the decrease of estradiol in the serum. The mechanism may be that the traditional Chinese medicines increased estradiol, causing the decrease of ALP, which induced the changes of Ca and P in serum, leading to a decreased excretion of Ca in urine. In this study, TJ-41 was effective in every parameter while TJ-7 and TJ-30 was effective on some parameters, showing that traditional Chinese medicines have specificities in the space environment. In conclusion, this study suggests that some traditional Chinese medicines may be beneficial for adaptation to a space environment.

The aim of this study was to evaluate the effects of herbal medicines in treating periodontal diseases. Three Chinese herbal composites [Conth Su (CS), Chi Tong Ning (CTN) and Xi Gua Shuang (XGS)], widely used for prevention and treatment of periodontal diseases, and the major components of these composites were tested for their ability to: (1) alleviate disease progression of experimental periodontitis in hamsters, (2) inhibit bacterial growth, and (3) induce mutations. Our results indicate that in treating experimental periodontitis, there were no significant differences between the animal groups with or without the use of Chinese herbal medicines in terms of the degree of inflammation, alveolar bone resorption, and rate of repair. However, hamsters treated with CS presented earlier regenerative epithelium. CTN demonstrated superior bacterial inhibition ability among all tested herbs (MIC 0.025 g/ml); CS showed good anti-bacterial abilities at a concentration of 0.05 g/ml. It is interesting to note that while both CS and CTN were capable of inhibiting bacterial growth, none of the individual herb components showed comparable bacterial inhibition abilities. None of the tested herbal composites or their components showed signs of inducing cell mutations using the Ames test. These results indicated that traditional Chinese herbal medicines, which have been used to treat periodontal diseases for hundreds of years by Chinese people, can effectively inhibit bacterial growth without causing cell mutation. Further investigation into their possible clinical applications in periodontal therapy is encouraged.

Eight weeks treatment with Unkei-to induced a significant increase in plasma follicle stimulating hormone (FSH), luteinizing hormone (LH) and estradiol levels in hyper- (robust) and hypo- (asthenia) functioning patients with first- and second-grade amenorrhea. We observed no significant differences in the rate of change of these hormones between hyper- and hypo-functioning patients. Ovulation occurred in 61.3% and 66.7% of patients with first-grade amenorrhea, and in 27.3% and 22.4% of patients with second-grade amenorrhea, respectively. No significant difference was observed in the ovulation rate after an 8-week treatment with Unkei-to between hyper- and hypo-functioning patients.

These results indicate that Unkei-to is effective in improving gonadotropin and estradiol secretion in the treatment of either hyper- or hypo-functioning anovulatory women.

There are conflicting records on the identity of Chinese medical herbs, and it is known that different plant materials are used under the same common names in different regions in China. However, there is no study on the genetic heterogeneity of medical herbs in any market outside of China. In this report, Chinese medical herbs under common names Radix Quinquefolii (American Ginseng or Xiyangshen), Radix Astragali (Huangqi), Radix Notoginseng (Tianqi), Coxtex Cinnamomum (Guipi), Radix Isatidis (Banlangen), Radix Codonopsis (Dangshen) and Radix Rehmannia (Shengdi) were collected from three independent herbal shops in Singapore and their DNAs were isolated and subjected to fluorescence Amplified Fragment Length Polymorphism (AFLP) analysis. While samples for Radix Quinquefolii and Radix Astragali were homogenous genetically [similarity index (SI) = 0.85 – 1.00] across the three shops, genetic heterogeneity was found for the other herbs (SI < 0.7). For example, four samples of Radix Codonopsis were of three distinct patterns (SI < 0.6). Our results highlight the situation that genetically distinct herbal materials are labeled and marketed under the same common names in an international market of Chinese medical herbs, which may contribute to inconsistency in quality and efficacy.

The purpose of this study was to evaluate the effectiveness of acupressure on gastrointestinal (GI) motility in women after trans-abdominal hysterectomy (TAH). Patients were randomly assigned into two groups of 21 and 20 patients each. The experimental group received acupressure for 3 minutes at each of three meridian points: Neiguan (PC-6), Zusanli (ST-36) and Sanyinjiao (SP-6). The control group received 3 minutes of acupressure on sham points. Acupressure was performed twice a day. A questionnaire was used to determine patients' satisfaction prior to and after afternoon acupressure. GI contractions were measured with a multifunctional stethoscope before and after acupressure. Acupressure of these three meridian points significantly (p < 0.05) increased GI motility in the experimental group, but there was little change in the control group (p > 0.05). Our conclusions are that non-invasive acupressure of these meridian points can significantly improve GI motility and can be incorporated into the technical curriculum and clinical education program of nursing schools. Patients and their family members can be taught to continue this procedure at home to enhance GI motility in patients who have undergone TAH.

The purpose of the present study was to investigate the relationship between the distribution of the analgesic area when different intensities of stimulation were applied to homotopic and heterotopic acupoints. The experiments were performed on volunteers; the left sural nerve was stimulated by a volley of eight rectangular pulses delivered at a frequency of 400 Hz. Electromyographic reflex responses (EMGs) were recorded from the ipsi-lateral biceps femoris muscle. Electroacupuncture stimuli were given on the skin overlying the Zusanli point (St 36), and the strengths chosen were times of the threshold eliciting a nociceptive reflex (T_RIII) response (0.6, 0.8, 1.0, T_RIII and T_supra-RIII, respectively). The effects of homotopic and heterotopic stimuli applied to St 36 on the pain sensation and the R_III reflex elicited by stimulation of the left sural nerve were observed to explore the pain-relief and R_III reflex-inhibition produced by stimulation of the St 36 point with different intensities. Both the nociceptive reflex and painful sensation evoked by stimulating the sural nerve were similarly inhibited by electroacupuncture at less than T_RIII applied to the ipsi-lateral acupoint. In other words, acupuncture with an innocuous intensity can produce homotopic pain-alleviating effects and reflex suppression. With contra-lateral electroacupuncture at the St 36 acupoint, innocuous intensities cannot produce heterotopic pain-relieving effects; these inhibitions were only observed at electroacupuncture intensities similar to the T_RIII threshold. These results suggest that local acupuncture-induced analgesia is effective with activation of large afferent fibers, whereas heterotopic acupuncture-induced analgesia is only effective with intensities strong enough to excite small afferent fibers. Local analgesic effects of acupoint stimulation involve segmental inhibition, whereas systemic analgesic effects of acupoint stimulation are involved in contra-lateral effects. The latter may recruit the diffuse noxious inhibitory controls (DNIC) system. The specific function of an acupoint is determined by the anatomical relationship between the disease focus and the segmental location of the acupoint.

The use of subjective end-points such as VAS pain scales in studies of acupuncture for chronic neck pain have resulted in equivocal results. This study introduces an objective parameter as the primary end-point for the assessment of acupuncture in patients with acute torticollis (stiff neck). Eighteen patients underwent a single 20-minute treatment session, with needling of two acupuncture points Hou Hsi (SI-3) and Luo Zhen (M-UE-24), on the side ipsilateral to the predominantly involved side of the neck. Measuring the angle of lateral head rotation using a simple compass and protractor, a mean improvement of 52.9% was found, more so among those presenting earlier (< 24 hours) as opposed to later (> 72 hours; p = 0.034). The use of objective parameters, as seen in acupuncture research of the gastrointestinal and respiratory tracts, should be incorporated into studies of acute and chronic neck pain. The use of sham needle points and placebo needles is problematic since both may elicit physiological responses.

Qigong is a complementary intervention for preventing and curing disease, and protecting and improving health through regulation of body and mind. Recently, we have been studying the psychoneuroimmunological effects of Qigong on the promotion of health. However, there are not many studies on the therapeutic efficacy of Qigong on various symptoms in Korea, hence the need to survey the clinical efficacy of Qigong. To evaluate the impact of Qigong in health care we categorized its effectiveness on the basis of ten years of subjects' memoranda. Among the 768 subjects, the motivation for doing Qigong was mostly to attend to health problems (81.5%), and males were more likely to use Qigong than females. The most improved symptoms were associated with psychological and musculoskeletal problems. Furthermore 66.9% of subjects reported improvements of perceived physical health and 40.3% of perceived psychological health. Other symptoms reduced by Qigong were pain (43.1%), fatigue (22.1%), and insomnia (8.7%). Wound healing was also surveyed (n = 332), and 84% of respondents reported improvement in recovery time, 66.6% reported reduced inflammation after Qigong and 50.3% reported no scarring as compared to before. In addition, 59.9% of respondents reported an increase in resistance to the common cold after four months of Qigong. The limitation of the study is that it is a retrospective survey on the basis of trainees' experiences of Qigong. Although this may constitute a potential bias, the study despite its limitations does provide precious empirical evidence of the effectiveness of Qigong.
