In this work, an attempt is made to analyze anatomical regions of Alzheimer's brain MR images using level set and Minkowski functionals (MFs). The T1 weighted sagittal view of normal and abnormal images considered in this work, are obtained from MIRIAD database. The ventricle along with the hippocampus is segmented using Reaction Diffusion (RD) level set method and is subjected to further analysis using MFs. The prominent feature derived from the segmented region which contains ventricle and hippocampus is correlated with the Mini-Mental State Examination (MMSE) score. Results show that RD method is able to delineate the exact boundary of the ventricle along with hippocampus. It is observed that the MF-area of segmented region provides better discrimination of normal and abnormal subjects (p < 0.001) compared to the MF-perimeter and MF-Euler number. The correlation of the MMSE with prominent MF-area for normal, mild, moderate and severe are found to be 0.71, 0.82, 0.84 and 0.89, respectively. This suggests that characterization of brain structures using MFs improve the discrimination capability of brain disorders. Hence MF-area feature could be used for the study of progression in Alzheimer's disease (AD) like neurodegenerative disorders.
Readhead et al. recently reported in Neuron the detection and association of human herpesviruses 6A (HHV6A) and 7 (HHV7) with Alzheimer’s disease by shotgun sequencing. I was skeptical of the specificity of their modified Viromescan bioinformatics method and subsequent analysis for numerous reasons. Using their supplementary data, the prevalence of variola virus, the etiological agent of the eradicated disease smallpox, can be calculated at 97.5% of their Mount Sinai Brain Bank dataset. Reanalysis of Readhead et al.’s data using highly sensitive and specific alternative methods finds no HHV7 reads in their samples; HHV6A reads were found in only 2 out of their top 15 samples sorted by reported HHV6A abundance. Finally, recreation of Readhead et al.’s modified Viromescan method identifies reasons for its low specificity.
Amyloidopathy is one of the most prominent hallmarks of Alzheimer’s disease (AD), the leading cause of dementia worldwide, and is characterized by the accumulation of amyloid plaques in the brain parenchyma. The plaques consist of abnormal deposits mainly composed of an aggregation-prone protein fragment, β-amyloid 1-40/1-42, into the extracellular matrix. Brillouin microspectroscopy is an all-optical contactless technique that is based on the interaction between visible light and longitudinal acoustic waves or phonons, giving access to the viscoelasticity of a sample on a subcellular scale. Here, we describe the first application of micromechanical mapping based on Brillouin scattering spectroscopy to probe the stiffness of individual amyloid plaques in the hippocampal part of the brain of a β-amyloid overexpressing transgenic mouse. Correlative analysis based on Brillouin and Raman microspectroscopy showed that amyloid plaques have a complex structure with a rigid core of β-pleated sheet conformation (β-amyloid) protein surrounded by a softer ring-shaped region richer in lipids and other protein conformations. These preliminary results give a new insight into the plaque biophysics and biomechanics, and a valuable contrast mechanism for the study and diagnosis of amyloidopathy.
