The hepatoprotective activity of Pinus massoniana bark extract (PMBE) against hydrogen peroxide (H₂O₂)-induced damage in normal human liver L-02 cells and carbon tetrachloride (CCl₄)-induced acute hepatotoxicity in mice was investigated. The L-02 cells were pre-treated with PMBE for 24 hours prior to exposure to 0.5 mM H₂O₂ for 3 or 24 hours. The cell viability, level of malondialdehyde (MDA) and glutathione (GSH), and the catalase (CAT) activity were evaluated. For in vivo experiments, mice were divided into groups and PMBE administered orally, after which each group was assigned a further treatment. Histopathological examination, the serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and GSH, the liver tissue levels of MDA and GSH, the activities of CAT and glutathione peroxidase (GSH-Px), were evaluated. PMBE treatment decreased the level of MDA and increased the cell viability, GSH content and CAT activity in H₂O₂ treated L-02 cells treated for 3 hours. PMBE obviously decreased serum ALT, AST, ALP, and liver tissue MDA, while increasing serum GSH, and liver tissue CAT and GSH-Px activities. In conclusion, PMBE treatment prevents H₂O₂ and CCl₄-induced liver damage, and therefore could have a potential clinical usage.

Calculus Bovis Sativus (also referred to as in vitro Cultured Calculus Bovis), an artificial substitute of natural Calculus Bovis (Niuhuang in Chinese, a traditional Chinese medicine), has been widely used to relieve fever, diminish inflammation and normalize gallbladder function in the last decade. This study aims to investigate the effects and possible mechanisms of Calculus Bovis Sativus on α-naphthylisothiocyanate (ANIT)-induced intrahepatic cholestasis in rats. Calculus Bovis Sativus (50, 100 and 200 mg/kg per day) was intragastrically (i.g.) given to experimental rats for seven consecutive days. A single dose of ANIT (100 mg/kg i.g.) was given to rats on the fifth day to induce intrahepatic cholestasis. The levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkalinephosphatase (ALP) and total bilirubin (TBIL) were determined through biochemical methods. The bile duct was cannulated with a PE 10 polyethylene tube to collect bile for two hours and bile flow was calculated by the weight of each specimen. Moreover, the mechanism of Calculus Bovis Sativus was elucidated by determining liver malondialdehyde (MDA) content and superoxide dismutase (SOD) activity. The biochemical observations were supplemented by histopathological examinations. Our results showed that Calculus Bovis Sativus (50, 100 and 200 mg/kg) significantly prevented ANIT-induced changes in bile flow and serum levels of ALT, AST, ALP and TBIL. Furthermore, Calculus Bovis Sativus (50, 100 and 200 mg/kg) significantly reduced the elevated hepatic MDA content induced by ANIT and increased the hepatic SOD activity suppressed by ANIT. Accordingly, histopathology of the liver tissue showed that pathological injuries were relieved after Calculus Bovis Sativus (50, 100 and 200 mg/kg) pretreatment. In conclusion, Calculus Bovis Sativus exerted a protective effect on ANIT-induced intrahepatic cholestasis in rats, which may result from the attenuated oxidative damage in liver tissues.

For centuries, Sophora alopecuroides L. has been used both as a food and an herbal medicine in northern China. A new cytisine-type alkaloid, N-methylene-(5,7,4${}^{\prime }$-trihydroxy)-isoflavone (LY01), was found in the fruits of Sophora alopecuroides L. and shows neuroprotective effects against Parkinson’s disease (PD). PD is a frequently occurring, irreversible neurodegenerative disease that seriously threatens the health of the elderly population. There is no cure for PD. The available treatments help manage the symptoms, but their use is limited by multiple side effects. Therefore, more pharmacological treatments addressing this pathology are urgently required. This study aimed to evaluate the neuroprotective effects of LY01 against PD, as well as their underlying mechanisms, using both in vitro and in vivo experimental models. A 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine hydrochloride (MPTP)-induced mouse model of PD was used to assess the effects of LY01 on the motor coordination deficit, progression of the pathology, and molecular characteristics. 1-Methyl-4-phenylpyridinium (MPP${}^{+}$)-activated SH-SY5Y cells and lipopolysaccharide (LPS)-activated BV-2 cells were used to evaluate LY01 effects on oxidative damage and neuroinflammation. In the rotarod test, LY01 alleviated the impaired motor coordination in PD mice. Furthermore, LY01 treatment prevented the loss of dopaminergic neurons in the substantia nigra and striatum of the PD mice, reduced neuroinflammation in the mice with MPTP-induced PD and the LPS-activated BV-2 cells, and diminished oxidative stress in the PD mice and the MPP${}^{+}$-induced SH-SY5Y cells. In conclusion, these results suggest the potential of LY01 as a therapeutic agent for treating PD.

This minireview describes the complexity of the molecular mechanisms involved in the tumor response to photodynamic treatment (PDT). Different aspects of reactive oxygen (ROS) and nitrogen species (RNS) induced by PDT will be examined. In particular, we will discuss the effect of ROS and RNS on cell compartments and the main mechanisms of cell death induced by the treatment. Moreover, we will also examine host defense mechanisms as well as resistance to PDT.

Objective: The aim of this study is finding an optimal and convenient aging marker.

Method: The information of participants’ occupation, family background, life style, physical condition, etc. was obtained by a self-administered questionnaire. Spot urine and saliva samples were collected during the early morning after fasting overnight. 8-hydroxydeoxygunaosine (8-OHdG), 8-iso-prostaglandin F2$\alpha$ (8-isoPGF2$\alpha$), advanced oxidation protein products (AOPPs), malondialdehyde (MDA), and dehydroepian drosterone sulfate (DHEA-S) were measured by ELISA.

Results: Around 112 non-smokers (males/females $=$ 61/51; mean age $=$ 44.9 years) were eligible and analyzed in this study. The mean urinary 8-OHdG level was $33.2\pm 1.75$ (ng/mmol, creatinine) and mean 8-isoPGF2$\alpha$ level was $364.97\pm 40.32$ (pg/mmoL, creatinine). In saliva, the mean level of AOPPs, MDA, and DHEA-S was $189.37\pm 19.81$ ($\mathrm{\mu }$mol/L), $1.7\pm 0.1$ ($\mathrm{\mu }$mol/L) and $2.96\pm 0.15$ (ng/mL), respectively. After analysis in multiple regression model, urinary 8-OHdG was influenced by age and tea consumption ($p<0.01$); urinary 8-isoPGF2$\alpha$ level was correlated with gender ($p<0.01$); salivary DHEA-S was related to gender ($p<0.05$) and marital status ($p<0.01$); AOPPs and MDA in saliva presented no relationships with those factors.

Conclusion: Among healthy non-smokers, urinary 8-OHdG is the best aging biomarker comparing to other markers.

Traditional Chinese Medicine (TCM) modernization has been proposed for many years, but the progress is still slow due to both ideological and technical obstacles. When I went to Japan in 1989, I found Japan has made a great progress on TCM by using modern technology. Therefore, I have studied a fine extract prepared from medicinal herbs (renamed Yi-Zhi-Yi-Shou, YZYS), a prescription of Dowager Cixi’s Yanling-Yishou-Dan of Qing Dynasty, with the current drug investigation strategies. I examined its antioxidant activity both in vitro and in vivo. The in-vitro studies found that YZYS possesses strong antioxidant capacity, such as scavenging various kinds of free radicals, and inhibits free radical-induced peroxidation of brain homogenate, microsomes, mitochondria, amino acids, deoxyribose and DNA. The in-vivo study with immobilization-induced emotional stress in rats, showed that YZYS effectively inhibits stress-induced stomach ulcers and oxidative damage in plasma and the brain. In addition, YZYS is shown to be non-toxic in both acute and chronic toxicity tests. These studies demonstrate that YZYS is a potent natural antioxidant and offer theoretical evidence for the beneficial effect of YZYS on health and brain functions, and that TCM prescriptions can be studied scientifically as modern medical drugs.
